Acceptors of fatty acid for glyceride synthesis in guinea pig mammary gland.

چکیده
T w o PATHWAYS have now been described for the biosynthesis of glycerides in mammalian tissues. In the pathway which exists in liver (1, 2) and adipose tissue (3), triglycerides are synthesized from L-a-glycerophosphate via L-a-phosphatidic acids and D-a,@-diglycerides. More recently it has been found that a second pathway involving the direct acylation of monoglyceride exists in intestinal mucosa (4-7), kidney (8), liver (8 ) , and aorta (9). During the course of our studies on fat metabolism in the lactating mammary gland (10-12), it became necessary to investigate the pathways of biosynthesis of glycerides in this tissue. Evidence will be presented in this paper for the existence in lactating mammary tissue of both known pathways of glyceride synthesis. Part of this work was presented at the April 1963 meeting of the Federation of American Societies for Experimental Biology (1 3).

Esterification of glycerol 3-phosphate in lactating guinea-pig mammary gland.
1. The presence of palmitoyl-CoA-l-glycerol 3-phosphate palmitoyltransferase (EC 2.3.1.15) has been demonstrated in a particulate fraction of mammary tissue from lactating guinea pigs. 2. Cell-free preparations also catalysed the activation of palmitate and oleate, and the conversion of enzymically formed phosphatidic acid into glycerides, in accord with the Kennedy pathway of glyceride formati...

3. Insulin did not alter the inactivity of rat mammary gland slices towards acetate as sole substrate, but in presence of glucose it considerably increased the 14C found in the fatty acids. The incorporation of "IC into the fatty acids of sheep udder slices wasunaffected by insulin both in acetate alone and in acetate plus glucose. 4. Glycerol caused a slight incorporation of acetate carbon int...
متن کاملThe effects of tannin-rich diet on protein composition of parotid and submandibular glands of Guinea Pig
Tannins are plant-derived polyphenolic compounds that are widely found in foods, particularly in fruits and beverages. These materials are water-soluble and able to precipitate proteins from aqueous solutions. It had been shown that feeding some animals such as rat and mouse with tannins mimicks the effects of isoproterenol on salivary glands. Tannins stimulate β-receptors by some indirect me...
متن کاملFurther observations on the in vitro stimulation by insulin of fat synthesis by lactating mammary gland slices.
The incorporation of [carboxy-14C] acetate into fatty acids by rat-liver slices in vitro was studied by Bloch & Kramer (1948), who showed that the rate of incorporation was increased by pyruvate and still further enhanced if insulin was present as well. The insulin effect was, however, not seen if glucose replaced pyruvate, nor did it occur in acetate alone. Later, Bloch (1948) reported that th...

1. Poly(A)-containing RNA was isolated from the nuclei of mammary gland, liver and brain of lactating guinea pigs. 2. Total nuclear poly(A)-containing RNA from mammary gland inhibited mRNA-directed protein synthesis by a wheat-germ cell-free system. It also inhibited the endogenous activity of the wheat-germ and other cell-free systems. It did not inhibit a wheat-germ cell-free system directed ...
